SenTraGor™ 

is an innovative reagent for the detection of senescence. Senescent cells play a potent role in normal development, tissue homeostasis, aging and a wide spectrum of age related disorders. Up to now, a major problem in the field of senescence was the absence of a reliable marker to specifically detect these cells, especially in situ. SenTraGor™ is a biotinylated Sudan Black B based chemical reagent that was designed to overcome this constraint.

It is the commercial brand of GL13 compound. 

 

This innovative compound is highly lipophilic and binds to lipofuscin, a hallmark of senescent cells. SenTraGor™ staining allows recognition of senescent cells in any biological material, including archival (Formalin Fixed Paraffin Embeded - FFPE), by visualizing cytoplasmic lipofuscin accumulation, via a hybrid histochemical/immunohistochemical assay. SenTraGor™ applications include: Immunohistochemistry (IHC), Immunocytochemistry (ICC), Immunofluorescence (IF), Flow Cytometry Analysis (FACS), co-Immunohistochemistry (co-IHC) and co-Immunocytochemistry (co-ICC). This new method is highly specific, sensitive and deprived of false positive and negative results.

IHC Protocol

The most innovative application of SenTraGor™ reagent is the clear cut detection of senescent cells in Formalin Fixed Paraffin Embbeded Tissues. Through classical immunohistochemistry, that can be applied in every pathology lab, senescent cells can be detected robustly in archive tissues.

co-IHC Protocol

You can study additional biological pathways and cellular markers in stained senescent cells from Formalin Fixed Paraffin Embbeded Tissues. The breakthrough technology of SenTraGor™ reagent allows the simultaneous detection of senescent cells and other cellular markers. Through classical immunohistochemistry senescent cells can be detected consistently in archive tissues along with standardized antibodies.

ICC Protocol

Staining of senescent cells with Immunocytochemistry is now performed easily by adding SenTraGor™ reagent before the primary antibody. Here are representative photos from stained senescent cells from different origin. Cells were incubated with SenTraGor™ before the addition of primary anti-biotin antibody and detection was performed with DAB.

co-ICC Protocol

One of the advantages of SenTraGor™ reagent is the ability to simultaneously detect senescent cells and a protein of choice from other biological pathways through Immunocytochemistry. After detecting the selected protein, proceed with the incubation of SenTraGor™ to discriminate the senescent cells.

IF Protocol

Immunofluorescence can be applied in biological specimens to detect senescent cells with SenTraGor™ reagent. Visualization of senescent cells with IF provides an advantage for concurently studying additional biological pathways inside those cells.

FACS Protocol

SenTraGor™ reagent can be used to detect senescent cells through Flow Cytometry Analysis. By simply incubating your preserved cells with SenTraGor™, subsequently using primary anti-biotin antibody and fluorescently labeled secondary antibody, senescent cells can be visualized.
